Dual GLP-1/Glucagon Receptor Agonist Weekly Protocol Guide

Survodutide Protocol Guide

✓ Verified dosing (fact-checked against FDA labels / published trials): Once-weekly subcutaneous injection. Initiate at 0.3 mg once weekly and titrate upward over ~14-16 weeks to a maintenance dose of 3.6 mg or 6.0 mg once weekly (Phase 3 obesity); MASH trials used 2.4/4.8/6.0 mg once weekly. Investigational only — not FDA-approved; no label-defined dose exists. This supersedes any conflicting figures shown elsewhere on this page; confirm with a licensed provider.

Survodutide is a dual GLP-1 and glucagon receptor agonist studied for obesity and metabolic dysfunction-associated steatohepatitis (MASH). The dual mechanism targets appetite suppression via GLP-1 while glucagon receptor activation increases energy expenditure and hepatic lipid oxidation.

Protocol Overview

Compound
Survodutide (Research Peptide for Obesity and MASH)
Category
Dual GLP-1/Glucagon Receptor Agonist
Vial Size
10mg (10,000mcg)
Reconstitution
2mL bacteriostatic water (5mg/mL)
Route
Subcutaneous injection
Frequency
Once weekly

Reconstitution Instructions

Using 2mL of bacteriostatic water results in a concentration of 5mg/mL. Start low and titrate gradually. Always use sterile techniques.

Materials Needed

  • 10mg Survodutide 10mg vial (lyophilized powder)
  • 2mL bacteriostatic water (sterile, preserved with 0.9% benzyl alcohol)
  • Sterile syringe and needle (18-22 gauge for drawing)
  • Alcohol swabs
  • Clean workspace

Step-by-Step Process

  1. Preparation: Allow the Survodutide 10mg vial and bacteriostatic water to reach room temperature. Wipe the rubber stoppers of both vials with an alcohol swab.
  2. Draw the Water: Using a sterile syringe, draw up exactly 2mL of bacteriostatic water. Expel any air bubbles.
  3. Inject into Vial: Insert the needle through the rubber stopper at a slight angle, aiming toward the glass wall. Slowly inject the water down the side of the vial to avoid foaming.
  4. Mix Gently: Remove the needle. Gently swirl or roll the vial between your hands. Do not shake vigorously.
  5. Check for Clarity: The solution should be clear once fully reconstituted. Do not use if cloudy.

Administration Guidelines

  • Administer subcutaneously once weekly on the same day each week
  • Start at 0.25mg weekly and titrate up every 4 weeks
  • Rotate injection sites between abdomen, thigh, and upper arm
  • Monitor for GI side effects during titration
  • Store reconstituted peptide refrigerated at 36-46F (2-8C)
  • Use reconstituted solution within 30 days
  • Never freeze reconstituted peptides
  • Document administration times and observations
